Kreirao ga je Sergio Bonelli koristeći pseudonim Guido Nolitta (iza kojeg je već stajalo nekoliko uspješnih serijala, Zagor prije svih), ali Mister No je imao malo dodirnih točaka sa ostalim Bonellijevim junacima: nije bio western, toliko omiljen čitateljima Texa, nije računao na zabavan i razuzdan mix načinjen od tisuću literarnih, filmskih i strip predložaka/polaznih točaka/varijacija na temu koji je krasio popularnog Zagora i definitvno se odricao vedrih i veselih postulata avanture na kojima je počivao uspjeh adolescentnih serija poput Kit Tellera ili Komandanta Marka. Tako na 8. stranici prvog broja na scenu stupa dinamičan i raspjevan mladić, koga život očito nije mazio (što je vidljivo po prerano posijedijelim zaliscima, borama smežuranom čelu i brazgotinama koje nosi po cijelom tijelu, uspomenama iz rata) i ulazi ravno među likove koji su ostavili dubok trag u priči o talijanskom stripu. On se zove Jerry Drake ali za sve nas on je jednostavno Mister No, nadimak koji je zaradio 1941. sudjelujući u američkom dobrovoljačkom odredu „Letećih tigrova“ koji su se borili na kineskoj strani protiv japanskog agresora (koji su između ostalog i zaslužni za to ime koje mu je dao okrutni pukovnik Saiko), nadimak kao stvoren za njega, koji će ga pratiti dok je živ! Mister No je uporno govorio NO (NE) japanskom pukovniku koji ga je ispitivao i mučio, NE krutoj vojničkoj hijerarhiji i užasima rata, NE licemjerju i lažnim vrijednostima suvremenog svijeta, NE tlačenju slabih od strane jakih! Mister No je Njujorčanin, veteran drugog svjetskog rata koji je sudjelovao u brojnim bitkama (u zračnim bitkama nad Filipinima, zatočen u logoru u Burmi, borio se na Guadalcanalu i Pacifičkim otocima, sudjelovao u oslobođenju Rima...) i koji se osjećao nesposobnim da se uklopi u normalan život nakon povratka iz rata. Zbog toga, u drugoj polovici 1950. odlazi u Južnu Ameriku, u Brazil i maleni gradić Manaus u potrazi za drugačijim životom neopterećen problemima suvremenog čovjeka, potragom za slavom i bogatstvom i pun prezira prema „american way of life“ i koji kao da je jedan od likova „On the Road“, knjige manifesta beat generacije Jacka Kerouaca i Neala Cassidya (s kojima se, nota bene, i susreće u jednoj od svojih brojnih avanutra), vođen jedino željom da živi slobodno s Piperom i bocom Whiskeya u ruci, ali upravo ga taj način života i poziv koji je odabrao prevozeći turiste diljem Južne Amerike uvlači u brojne nevolje i prisiljava da pribjegne nasilju od kojeg je bježao i koje ga poput sjene prati gdje god krene. Mister No je, kako je sam Sergio Bonelli izjavio u jednom interwievu, nastao prema stvarnom liku kojeg je Sergio vidio na jednom od svojih brojnih putovanja:“...Da, bio sam u Palenqueu, u Meksiku i susreo sam mladog i zgodnog mladića na ruševnom aerodromu.Nosio je kožnu jaknu i zvao se Kapetan Vega, ime koje je perfektno odgovaralo strip junaku! Imao je opasač s pištoljem i letio je po Yucatanu gore dolje prevozeći životinje, povrće i razne namirnice i kad god je sreo turiste, samo bi pokazao na sjedalo za putnike i rekao:`Ok, odvest ću vas gdje god želite`! Tada mi se rodila ideja o junaku koji će biti pilot Pipera, ali sam odlučio serijal smjestiti u Amazoniju....“ Mister No je inače svojevrsni alter ego svog oca, Sergia Bonellija, lik kroz koji Sergio progovara o svojim velikim ljubavima osim stripa: jazz glazbi, putovanjima i velikim prostranstvima, moralnim problemima suvremenog društva, a nerijetko su avanture opisane u njegovim epizodama inspirirane dugim posjetima Amazoniji, gdje bi Sergio išao u potrazi za inspiracijom, ali i Africi i drugim mjestima gdje ga je neobuzdani pustolovni duh odveo! Mister Noova „sadašnjost“ je kraj 50-ih godina prošlog stoljeća, ali njegova „biografija“ je potanko objašnjena tijekom izlaženja, počevši od najranijeg djetinjstva i ranih 30-tih kada je kao mali dječak odrastao na vrelom asfaltu Brooklynških ulica preko ratnih 40-tih i ratišta širom svijeta sve do dolaska u Brazil. Većinu vremena Mister No provodi ljenčareći po Manausu ispijajući bezbrojne boce kačake i viskija sa svojim prijateljima u konstantnoj potrazi za mušterijima, bili oni inženjeri, misionari, bogati turisti, istraživači ili naučnici. Vjerni pratitelj u mnogo avantura mu je Otto Kruger ili Esse Esse, bivši pripadnik njemčke SS divizije čija je prošlost dijelom objašnjena u tri kratke priče objavljene kao dodaci specijalnim brojevima. Esse Esse je bivši njemački vojnik ali ne i nacist jer je bio primoran da sudjeluje u ratu mimo svog pristanka. Kruger je ekspert u rukovanju nožem i nerijetko je koristeći ga spasio Mister Noa i sebe iz naizgled bezizlaznih situacija. Zatim, jedan u nizu važnih Mister Noovih prijatelja je Phil Mulligan, bivši suborac na Pacifiku i privatni detektiv koji je postao pisac koji pše kriminalističke priče u kojima je glavni junak niko drugi nego Jerry Drake! U Manausu imamo nezaobilaznog Paola Adolfa, simpatičnog barmena koji vječno utjeruje dug od Mister Noa, pa simpatični crnac Dana Winter, pjevač i gitarist, Narednik Oliveira koji uporno sanja dan kad će spremiti Mister Noa iza rešetaka i cijelu plejadu mehaničara, barmena, pilota,policajaca i muzičara širom Južne Amerike. Važnu ulogu u Mister Noovom životu zauzimaju žene! Mister No je okorjeli zavodnik, pravi Don Juan sa puno uspjeha kod žena, ali koji uporno izbjegava zamku braka u koju su ga moge neuspješno pokušale namamiti (uz jednu iznimku, Mister No je proveo jedan dio života kao pripadnik plemena Yanoama i bio oženjen prelijepom Indios djevojkom koju je ubio ljubomorni suplemenik). Svakako, najvažnija od svih žena u njegovom životu je lijepa plavokosa profesorica arheologije Patricia Rowlands s kojom je imao nekoliko avantura, zatim lijepa pilotkinja Debora, pa strastvena Miranda Cordeiro, kćerka jednog od zemljoradnika iz Sertaoa, pa tri lijepe vlasnice Ipanema bara u Manausu, Madalena, Irene i Maria, tajna agentica Delia... Iako se serijal odvija u ranim pedesetim godinama, žene koje Mister No sreće uglavnom su odlučne i nezavisne moderne žene i uglavnom igraju pozitivnu rolu u njegovim avanturama, mada smo sreli i par negativki! Mister No nema arhetipskog neprijatelja koji se opetovano pojavljuje kroz cijelu sagu. Njegovi neprijatelji su obično prikazani kao naizgled normalni ljudi koji tek nakon što ih Mister No preveze na željeno mjesto pokažu svoje pravo lice i ambicije, jedna široka galerija likova koja nije prikazan u crno-bijelom tonu koji dozvoljava čitatelju da donekle i simpatizira sa negativcem.Ipak, jedan od najupečatljivijih negativaca je japanac Ishikawa, u neku ruku nemilosrdni alter ego Mister Noa, kojeg shvaćanje i osjećaj časti tjeraju da uništi Jerryjem život i s kojim se Mister No obračunava u borbi na život i smrt na New Yorkškim dokovima. Sergio Bonelli je dugo bio glavni scenarist Mister Noovih pustolovina da bi konačno krajem osamdesetih napustio pisanje scenarija pritisnutim obvezama vođenja izdvačke kuće, a glavnu ulogu je preuzeo Luigi Mignacco. na stvaranju Mister Noa velik obol su dali i ostali poznati scenaristi kao Alfredo Castelli, autor Martin Mysterea, Tiziano Sclavi, autor Dylan Doga, Maurizio Colombo, Stefano Marzorati, Michele Masiero, dok velike zasluge za grafičku relizaciju lika nosi Roberto Diso koje je redizajnirao lik i podario mu svoje lice (lik Mister Noa je praktički autoportret mladog Roberta Disoa) i tako definirao Mister Noa kakvog ga znamo danas, iako je Gallieno Ferri crtao svoju, originalnu verziju Mister Noa na naslovnicama. Mister No je nedavno proslavio 300. broj, ali je serijal u dubokoj krizi i samo ga činjenica što je Sergio Bonelli emotivno vezan za svog „sina“ spašava od zatvaranja.
